First, press the green flag. Then there are 5 levels of difficulty to choose from. Baby Mode, Easy, Standard, Hard, and Impossible. (There's also Sandbox mode!) Once you've chosen your difficulty, you earn points constantly. You use points to buy bugs that try to fight through the opposing CPU's bugs and get to their tree to damage it. However, you also have a tree to defend. Try to defeat your opponent before they can defeat you!
